New Super Mario Bros. (DS cartridge) has no in-game save system on DSi. The game only autosaves after completing a castle/fortress or using a Star Coin to unlock a path. You cannot manually save mid-level.
Finish a level (reach the flagpole) before suspending. That completion is saved to the cartridge. Mid-level suspend is a convenience, not a save.
